How do I solve the missfiring on one cylinder?...

I have checked thee plug and HTcable and they are both working, and yet it is still misfiring on the one cylinder which I believe is #4. It is a vr6 that I recently purchased. It was running ok when all of a sudden itt began to miss fire.
